蓝色葡聚糖以受控的葡聚糖片段为原料，与Cibacron Blue F3GA反应合成得到，经非结合染料纯化后，对产物的平均分子量、溶解度、取代度、游离染料和干燥失重进行了控制。低分子量蓝色葡聚糖（BD5、BD10和BD20）在生产过程中需要通过色谱法进行纯化，因此比大分子量的蓝色葡聚糖更昂贵。TdB实验室提供各种分子量的蓝色葡聚糖，范围从5 kDa到2000 kDa。

合成与结构

葡聚糖来自肠系膜明串珠菌B-512F，其本质上是一个线性的α-（1-6）-连接的葡萄糖链，沿链分布的α-（1-3）支链百分比较低（2-5%）。所使用的葡聚糖组分的重量平均分子量（Mw）为4000到2000000。葡聚糖用Cibacron Blue F3GA标记合成蓝色葡聚糖。

产品由葡聚糖的近似分子量表征。BD5具有约5000的分子量。通过凝胶色谱（GPC）测定的实际分子量来确定。该值随分析证书一起提供。

物理性质

蓝色葡聚糖是一种蓝色、无味粉末，极易溶解于水或电解质溶液。由于Cibacron Blue结构中同时存在还酸盐和氨基，该产品具有显著的两亲性特征。由于这种特性会影响溶液的流体动力学体积以及与凝胶的相互作用，因此在GPC检测样品时会发现一些异常。蓝色葡聚糖不溶于大多数有机溶剂，例如乙醇、甲醇、丙酮、氯仿、乙酸乙酯等等。图.2蓝色葡聚糖110水溶液的吸收光谱。蓝色所色团的吸光度max为621.5nm（见图2）

稳定性

蓝色葡聚糖稳定性尚未有前瞻性的研究。然而，对室温下3年蓝色葡聚糖样品未显示出不稳定。建议将产品储存在密封容器中，避光。蓝色葡聚糖可在室温下储存。蓝色葡聚糖在室温下干燥储存在密封良好的容器中可稳定6年以上

毒性

Haz-Map数据库中蓝色葡聚糖被归为皮肤、眼睛和呼吸道刺激物。建议与大多数粉末一样，应避免吸入，尤其是反复接触。

应用

长期以来蓝葡聚糖2000一直用作色谱分析的空白体积标记，蓝色葡聚糖凝胶共轭物用于色谱分析也已经许多年（参见GE Healthcare Life Sciences；尺寸排除色谱法；原理和方法）。现在TdB提供的蓝色葡聚糖拓展了生物科学中的应用范围，且可使用标准UV和RI检测器进行检测。蓝葡聚糖已经用于其他重要的研究领域，如下所列：溶酶体活性研究（1）、内皮细胞通透渗透性（2）、牛精子渗透性y（3）、角膜渗透性（4）、肺血流研究（5-7）、大脑渗透性（8-10）、蛋白质和酶与蓝色葡聚糖的结合（11-13）。
